About 200 of the mice, each weighing the same as a 10 pence coin, have been released into the Northumberland Wildlife Trust's East Chevington Preserve in Druridge Bay.
This is the second time they have been introduced to the area - 17 years ago 240 were released and the colony has survived to this day.
Their release was made possible by a large private donation.
It will strengthen what is one of the few Western Harvest Mouse colonies north of the River Tyne.
Duncan Hutt, the trust's conservation manager, said it was "fantastic" to see the mice come out on the spot.
n 2004, Wendy Fall, a doctoral student at the University of Newcastle, actively raised 240 harvest mice with the help of volunteers, schools and community groups.
The mice, which are Britain's smallest mammal, have been released to the 185-hectare (457-acre) reserve as the vast reed beds provide suitable habitat.
Animals are expected to build distinctive circular grass nests on tall plants, three feet above the ground.
